Cellular imaging by electron microscopy

Introduction

Different techniques are available in the platform to allow the observation of biological samples

1. Fixation

  • Chemical fixation
  • Fixation by high-pressure freezing

2. Inclusion

  • Classical embedding (EPON, LR White)
  • Cryo-substitution and embedding

3. Sectioning

  • Classical sectioning
  • Cryo-sectioning (Tokuyasu)

4. Labeling and Staining

  • Post-staining
  • Immunolabeling (manual or automated)

5. Observation

  • Trasmission Electron Microscopy

Sample types

Cell monolayer or pellet, bacteria, or tissue.
Samples have to be at the biohazard level L1.
L2 samples must be chemically fixed before.

Results

The results allow:

  • Morphological studies of cells and bacteria
  • Protein localization at a sub-cellular level
  • Determination of virus infection cycle

Development of correlative microscopy is in progress.

Data Storage

Data are stored for 3 years.
