CM02 is a Titan Krios G4 (Thermo Fisher Scientific), equipped with a cold FEG, a Selectris X energy filter and a Falcon 4i electron detector. It is operated as an ESRF CRG beamline: one third of the beam time is allocated to ESRF users while the rest is available for French laboratories through a curated process. This cryo-electron microscope is able to carry out single-particle and tomography experiments. CM02 is part of the « France Cryo-EM » infrastructure financed by the “Plan Investissement d’Avenir 3” scheme which has enabled the purchase of 3 state-of-the-art 300 kV cryo-electron microscopes in France. It is maintained and operated by a team of 5 people from the IBS/ISBG (Grenoble).Technical specification

Cost
Microscope time is free of charge for 2025. All other expenses are the user’s responsibilities.
ESRF access (one third of total beam time)
Applications to use CM01 and CM02 Krios microscopes are made through the ESRF User Portal (SMIS). The type of proposal is a structural biology rolling-access application, which can be submitted at any time during the year. Additionally, specific cryo-EM BAGs have been created (which are distinct from the MX-BAGs). Contact: cryo-em@esrf.fr
Cost
For accepted proposals, travel expenses will be reimbursed for a maximum of two visitors per experiment.

Sample requirement
Evidence of sample quality and suitability must be given to justify the use of this Titan Krios microscope.
- For SPA experiments, include results such as typical micrograph, 2D class averages, 3D reconstruction, atlas, etc.
- For molecular cryo-ET: Atlas and high magnification image of the sample.
- For cellular cryo-ET: Atlas and medium magnification images showing cells/lamellae with the regions of interest.
Grids must have been screened and clipped in autogrids compatible with the autoloader system. Up to 4 grids will be loaded on the microscope.
Experiments
Single particle analysis
Typical SPA sessions are 48h. On demand, 24h sessions can be allocated. In specific cases, sessions up to 72h can be offered if justified.
Several grids can be collected in one session with the multigrid acquisition strategy, with the same data collection parameters.
Tilted acquisition (up to 40°) can be considered if previous high resolution data acquisition analysis displayed strong preferential orientations not solved by sample preparation optimization.
Tomography
Tomography sessions can be of 48-72h depending on the needs.
