Analytical ultracentrifugation (AUC) studies macromolecules in solution that are subjected to a centrifugal field. AUC combines the separation of the macromolecules and the analysis of their transportation in view of a rigorous thermodynamics.
Using interference and absorbance detection, or fluorescence detection, Sedimentation velocity and Sedimentation equilibrium experiment can be done to characterize different macromolecules as proteins, nucleic acids, lipids, polysaccharids, AAVs, nanoparticles …
Analytical ultracentrifugation is particularly suitable for studying the interactions of membrane proteins, glycosylated proteins, complexes between proteins and polymer or RNA…
Application
- Determination of aggregation and oligomerisation state: homogeneity of the sample
- Determination of general shape of the macromolecule: molecular mass, hydrodynamic radius
- Determination of the association state of complexes
- Determination of the association constants and weak inter-molecular interactions that determine protein solubility
- Determination of the stoichiometry of multi-component systems, on the basis of their density and optical properties (absorbance, refractive index and fluorescence).

How to make a request ?
This instrument works in service mode.
The applicant should fill in the first part, which is completed in agreement by both part, in order to define the best way to conduct the experiments, before signature of the parts. Each new project manager should contact us, sending by e-mail the request form (with the part 1- and 2- filled).
After discussion on the project, the part 3- (experimental conditions) is filled and the experiment planned.
Then, you accept the experimental conditions, defined in the request form, and the general terms.
Samples
All information are given in the condition of use for AUC.
Storage conditions of samples will be defined, by the applicant, in the request form.
